Clinical Pharmacology
Griseofulvin is fungistatic with in-vitro activity against various species of Microsporum, Epidermophyton, and Trichophyton. It is deposited in keratin precursor cells and is concentrated in the stratum corneum of the skin and in the nails and hair, thus preventing fungal invasion of newly formed cells. Griseofulvin is metabolized by the liver mainly to 6- dimethyl griseofulvin which is excreted in the urine.

The drug is contraindicated in patients with prophyria, hepatocellular failure, and in individuals with a history of hypersensitivity to griseofulvin. It may diminish the effects of coumarin anti coagulants and oral contraceptive possibly by increasing their rate of metabolism. It may enhance the effects of alcohol. Effects of griseofulvin is diminished by concurrent therapy with barbiturates. Use of griseofulvin in pregnancy is not recommended.
